Factors responsible for pig infections caused by the significant respiratory pathogens porcine circovirus type 2 (PCV2), porcine reproductive and respiratory syndrome virus (PPRSv), and Mycoplasma hyopneumoniae (M. hyopneumoniae) were investigated in a cross-sectional study. In Uganda, the presence of hyo, Actinobacillus pleuropneumoniae (App), and gastrointestinal (GI) parasites is a significant concern. To gather data on infection-related management methods, a structured questionnaire was used. A survey was conducted on 90 farms, encompassing a total of 259 pigs. Sera samples were screened for the presence of four pathogens using commercially available ELISA assays. The Baerman's method was used to characterize parasite species found in faecal samples. Identifying infection risk factors involved employing logistic regression. Individual animal serological prevalence of PCV2 demonstrated a value of 69% (confidence interval 37-111). For PRRSv, a seroprevalence of 138% (95% confidence interval 88-196) was found, along with 64% (95% confidence interval 35-105) for M. hyo, and a strikingly high 304% (95% confidence interval 248-365) for App. A notable prevalence of Ascaris spp. was observed at 127% (95% confidence interval 86-168), coupled with a high prevalence of Strongyles spp. at 162% (95% confidence interval 117-207), and an extremely high prevalence of Eimeria spp. at 564% (95% confidence interval 503-624). The onchocercid nematodes of the subfamilies Dirofilariinae and Onchocercinae exhibit a required mutualistic interdependence with Wolbachia. For the intracellular bacterium found in the filarioid host, in vitro cultivation has not yet been attempted. As a result, the current study employed a co-culture system of embryonic Drosophila S2 cells and LD cell lines for the purpose of cultivating Wolbachia from Dirofilaria immitis microfilariae (mfs) harvested from affected dogs. Shell vials, augmented with Schneider medium, served as the inoculation sites for 1500 microfilariae (mfs), employing both cell lines. During the initial inoculation, and at subsequent medium changes from days 14 to 115, the multiplication and establishment of the bacterium were under observation, commencing at day zero. A 50-liter aliquot per time point was examined by quantitative real-time PCR (qPCR). In evaluating the average Ct values from various parameters, including LD/S2 cell lines and mfs with and without treatment, the S2 cell line lacking mechanical disruption to the mfs showed the highest quantifiable Wolbachia count by qPCR. Sustaining Wolbachia in co-cultures derived from both S2 and LD cells for 115 days, while promising, still leaves a definitive conclusion far off. To elucidate Wolbachia infection and cell viability in the cell line, further experiments using fluorescent microscopy and vital staining are necessary. To improve infection susceptibility and develop a filarioid-based cell line system, future investigations should utilize a considerable quantity of untreated mfs to inoculate Drosophilia S2 cell lines and include the addition of growth stimulants or pre-treated cells to the culture media.
